New primer sets for early bovine leukemia virus (BLV) detection are designed on the basis multiple alignment of gene env sequences of different BLV isolates from EMBL/ GenBank and following thermody- namical analysis of primers and amplicons. Theoretical and experi- mental comparison of designed and known primer sets for BLV de- tection is carried out. It is shown designed primers have higher spe- cificity and accuracy than known primers. Main principles of polyme- rase chain reaction (PCR) test-system characterization and primers design are modified. It is shown primers homology degree is a key parameter for PCR accuracy. The universal primer system for preim- plantational cattle, sheep, goat and pig embryo sexing is created.
