Essay

Dissertation is devoted to investigation of influence of new fluorine-containing activators of KATP channels on heart function and vasomotor reactions under physiological and pathological conditions. Using specific inhibitor - glibenclamide - we showed that new agents really act as activators of this channels. The tested agents showed tremselves powerfui vasodilators;their effects were dose-dependent and dependent on the initial vascular tone. Norepinephrine and, especially, angiotensine II partially inhibited their vasodilator effects; i.e., it can be supposed that the above agents suppres the KATP channel activity. Angiotensin II-induced inhibition of their effects was stronger in preparations from hypertensive rats. We should mention that dilatory effects of new activators of potassium channels tested in normotensive and hypertensive preparations under conditions of K-depolarization were rather similar. PF-10 enhanced myocardial contractility and attenuate spontaneous alterations of the heart rate.Preliminary activation of KATP channels by PF-10 exerted a cardioprotective effect under cjnditions of ischemia-reperfusion (similary to the well-known phenomenon of ischemic preconditioning).
