Essay

Dissertation is devoted to working out the model system of the process of biotechnological acetaldehyde production. Ethanol into acetaldehyde conversion by alcohol oxidase of methylotrophic yeast Hansenula polymorpha mutant cells was studied. The influence of mutations which block the activities of acetaldehyde metabolism enzymes, affect the glucose repression of alcohol oxidase synthesis and ethanol-induced alcohol oxidase inactivation on efficiency of ethanol to acetaldehyde biotransformation was investigated. The new method for isolation of mutants with high levels of alcohol oxidase was elaborated. The permeabilized by digitonin cells of regulatory mutant strain with high level of alcohol oxidase under optimal conditions was converted 96,4% of ethanol into acetaldehyde in presence of the vacuolar proteinase inhibitor phenylmethylsulfonyl fluoride.
