Essay

The work is dedicated to studying the effect of temperature and osmotic medium parameters, as well as of modificators of a cytoskeleton-membrane complex, amphiphilic compounds and inhibitors of anion transport on erythrocytes susceptibility to hypertonic shock and hypertonic cryohaemolysis. Chemical modification of erythrocytes was performed using cells treatment with hemin and diamide, such amphiphilic compounds as chlorpromasin, trifluorperasin, haloperidol, dodecyl-beta-,D-maltoside, hexyl-beta,D-glucopyranoside, octyl-beta-D-glucopiranoside, decyl-beta, D-glucopyranoside. Ionic transport modification was carried-out by anion transport inhibitors: DIDS, SITS, furosemide, salicylic acid, dipiridamol. The medium composition was varied with by adding penetrating cryoprotectants (DMSO, glycerol). Obtained results testify that erythrocytes stability to hypertonic shock and hypertonic cryohaemolysis can be increased by using the certain cell regimens modification using cryoprotectants, modifiers of a cytoskeleton-membrane complex, as well as anion transport inhibitors. In this case the temperature and medium osmolarity are known to be additional parameters, the use of those can directly increase the cell stability.
