Essay

The thesis is concerned to the development of the approach to the exposure of the viruses directly in cell homogenates using specific antibodies by the surface plasmon resonance method. The new modification of the sensor surface for the detection of visusspecific macromolecules by forming of the monomolecular layer with the effective negative charge at it's surface is proposed. This modification prevents proteins denaturation and provides their oriented immobilization. The general approach to the detection and quantitative determination of viruses by SPR method using specific antibodies, immobilized at the sensor surface modified by KNCS and protein A St. aureus is developed and tested by the example of tobacco mosaic virus. By the example of green algae Bracteаcoccus minor cells infected at the zoospore stage, it was shown, that such a modification of the sensor surface allows adequate detaction of the virus in cell homogenates. It was confirmed by IFA and light microscopy methods. The possibility ofthe use of this approach for viral proteins was shown on the example of the structure protein of potato virus X and hexon of adenovirus 2. The new original procedure of the quantitative determination of the virus by immobilization of the complex specific IgG-virus at the KNCS and protein A-modified surface was proposed. With the example of Nicotiana tabacum the development of the viral infection in the plant was studied by the mentioned approach. The agreement between SPR and ELISA methods was achieved.
