Essay

Inhibitors of calf intestine alkaline phosphatase (AP). The studying of kinetic regularities of calf intestine alkaline phosphatase inhibition by L-cysteine, reduced glutathione and dihydrolipoic acid - and also to creation of new effective organophosphorus inhibitors of this enzyme. Kinetics, spectrophotometry. It was shown, that effect of reduced glutathione on activity of AP is described by partial mixed-type inhibition. The mechanism of equilibrium establishment between L-cysteine and enzyme involves the initial rapid formation of intermediate complex and a subsequent slower step leading to its stabilization at the substrate binding site. It was shown, that a-amino- and b-aminophosphonic acids can be the activators AP. The sphere of use - bioorganic chemistry, biochemistry.
