Morozova I. The cryopreservation of mouse embryo at high and super high freezing-thawing rates

The thesis is devoted to the investigating of the effective ways of realisation high and super high freezing-thawing rates of embryos. The complex comparative estimation of different ways of realisation high and super high freezing-thawing rates was spent. The device (patent №98041888) for cryopreservation of cell suspension at super high freezing-thawing rates is developed in collaboration with other scientists. The dependence of the solution turbidity degree on concentration of glycerol and sucrose and the freezing-thawing rates is established. The mathematical formula for definition of the minimal concentration cryoprotectant wich provides the vitrification of solution at the set rates is received. The influence on the survival level of mouse embryos at cryopreservation by means high and super high freezing-thawing rates of such factors as ratio of thawing rates to rates of freezing, volumetric expansion of cryoprotectant solution environment at freezing, exposition terms of mouse embryos in cryoprotective solutions before freezing is studied. A method of mouse embryo cryopreservation, basing on application of closed straw with diameter of 1,0 mm, is offered.
