Research subjects: morphogenetically processes in protoplasts culture of white-head cabbage in vitro; Research objectives: to reveal stage of white-head cabbage morphogenesis in culture of protoplasts in vitro and to improve biotechnology of white-head cabbage micropropaganation; Methods: biotechnological, cytological, biometric, biochemical and molecular-genetic ones; Theoretical and practical results: for white-head cabbage varieties of native breeding (Brassica L. oleracea var. capitata L.) for the first time there are determinated regularities of morphogenetic processes in protoplasts culture and on their basis there is worked out the system of plant regeneration in vitro. For realization of morphogenetic potential of cells and tissues for the first time there is shown regulating activity of new chemical compounds derivated from phenyl-acetic acid (DPGH-1, DG-036, DG-048 and DG-049) and pyridine (DG-466, DG-482 and DG-735). In the result of conducted investigations there are revealed new regulatorsof dedifferential, cytokinin and osmoprotector action. Regulating role of gibberellin (GK3) in determination of cells differential division in protoplasts culture of 6 species of plant from Brassicacea and Solanaceae family is confirmed. The method of genetic differentiation of white-head cabbage varietal material for selection of initial foerms, when creating high-heterozygous F1 hybrids, is offered; Novelty of introduced: on the basis of worked out biotechnology of cells growing there is received plants regeneration in protoplasts culture of 6 white-head cabbage varieties, breeded at Institute of Vegetable and Melons UAAS - Kharkivs'ka Zimova, Ukrains'ka osin', Yaroslavna, Lesya, Lika, Bilosnizhka and one variety of Russian breeding - Iuns'ka Rannya, this gives the possibility to use this varietal material in cell-engineering technology. A new variant of the scheme for cell breeding of heterokaryons is worked out. There are received intervarietal somatic hybrids of white-head cabbage in combinations of protoplasts of white-head cabbage variety Ditmarsher Fruer with the varieties Kharkivs'ka Zimova, Ukrains'ka osin' and Yaroslavna. The system of isoferments markers (on the basis of esterase and aspartataminotransferase) is selected, as well as 2 RAPD-primers, suitable for evaluation of white-head cabbage intervariental polymorphism, are revealed. For the needs of plants biotechnology there are studied and introduced in practice new regulators of dedifferential, cytokinin and osmoprotector action, offered new gelforming preparations of nutrient medium; Implementation degree: by the results of conducted investigations there are received 3 licences and 2 patent application for an invention. The thesis data were used in breeding work, when creating heterotic hybrids at IVM UAAS. In 2003 in the result of testing of 17 variety-linear hybrids, created on the basis of varieties, breeded at the Institute, there are received forms, which have excess by the yield over the standard about 9,7 - 22,2 t/ha; Effectiveness: thedetermined optimal tropic, osmotic and hormone factors of growth and differentiation of cells in protoplasts culture in vitro permit to reproduce stably plants regeneration by organogenesis of different white-head cabbage varietal genotypes; Sphere, sector of implementation: agricultural biotechnology, training of specialists.
