Essay

The thesis is dedicated to studying the effect of low positive temperatures on spermatozoa fraction with a fast progressive movement, isolated from human ejaculater on its morphofunctional characteristics. In order to solve the tasks set in the work the technique for storage of actively motile fraction of spermatozoa, isolated from human ejaculate was elaborated. When selecting temperature and storage medium the highest integrity of kinetic activity, gamete viability and acrosin activity was observed under 8°C in the Menezo B2 medium. According to the investigations results the spermatozoa, stored under hypothermia conditions within 24 hrs were shown to preserve their fertilising ability. Proceeding from these data there was elaborated the technique for insemination of immature oocytes after their maturation and for reinsemination of non-fertilised oocytes with spermatozoa, stored under hypothermia has been elaborated. The carried-out investigations testify to the efficiency of this technique performance, that allows to increase the frequency of fertilisation and to augment the pregnancy percentage.
