Essay

The level of virus infection on grapevine cultivars and clones is studied. The high level of grapevine fleck virus infection is showed. The amount of grapevine fanleaf and leafroll virus infection is not so high. It has been demonstrated that clonal selection together with sanitary selection is a very effective method to decrease the level of virus infection. The optimization of ELISA-test for three grapevine viruses is done. The best tissues and organs for reliable ELISA detection is estimated. Planting material from USA is tested for nepoviruses. DsRNA from grapevine affected by vein mosaic, rugose wood and grapevine leafroll is characterized. The method of grapevine latent infection detection by hybridization with radioactive labeled probe (dsRNA associated with leafroll) was elaborated. The sensivity of this method is near 1 - 10 ng of virus RNA per sample. The distribution of nematode species on clonal assays plots is studied. It has been established that the main species of coccides in Odessa region vineyards is Pulvinaria vitis, Parthenolecanium corni and Pseudococcus longispinus. The last one (as is shown by our experiments) can transmit grapevine leafroll virus. By thermotherapy and meristeme culture methods the healthy initial material of Golubok and Tairov's Rubi is obtained.
