Essay

The dissertation is dedicated to studying of fundamental biological properties and energy processes wich take place on the membrane of gonococci resistance to antibiotics. The results of the main parameters of the gonococci strains growth are given this work, the principal factors of pathogenicity were explored, the basic sources of the converted energy of the reseached strains were defined, generation protonmotive force was studied, ATPase activity and the content of the intracellular ATP were determined. The optimum composition of the culture medium for cultivation N. gonorrhoeae with 20% content of cattle serum (CS) was selected and it was determined that the optimum development of the reseaching strains occures when pH = 6,8-6,9 the temperature is 36,5oC-37,5oC. The dependence on the factors of pathogenicity in the reseached strains gonococci from resistance to antibiotics was shown. For the first time it was determined that there are two main sources of the converted energy in the cells of the reseached strains: transmembrane difference of the electrochemical potentials and intracellular pool ATP. It was shoun that the brining oh the difference of the electrical potential or gradient of pH on the membrane of gonococci induce the synthesis ATP, formation of the ionic gradients. The content of ATP in the gonococcus cells is in limit from 0,053 till 0,064 mM. For the first time it was determined the magnitude of the protonmotive force (183-192 mv) cells N. gonorrhoeae and its components: transmembrane difference of the electrical potentials (103-145 mv) and concentration gradient of protons (80-47 mv). Transmembrane electrophoresis of the penetrative ions TPP+ and PCB- showed that the membrans of the intact cells and the membranes of the gonococcus vesicles contain theree types of generators of the membrane's potential: respiratory chain, ATPase and transhydrogenase. It was determined for the first time that the reseched strains Neisseria gonorrhoeae resistance to antibiotics have ATPase activity 2,0-2,5 higher than sensitive it can attest more intensive energetic metabolism.
