Essay

Thesis is dedicated to the study of energetic state of liver during cold storage and normothermic reperfusion. It has been shown that sucrose-based solution (SBS) is able to preserve ATP level of liver and does not yield to commonly accepted University of Wisconsin solution (UW). 1-hour cold storage in the UW causes accumulation of potassium ion in mitochondria, which is retained during 24-hour cold storage. Normothermic reperfusion after 24-hours equalizes the difference between two solutions because of mitochondrial potassium ion release in the UW-preserved livers. ATP-synthase inhibition and increase of olygomycin-insensitive activity are the main reasons of respiration rate decrease after 24-hour cold storage in the two preservation solutions. Albumin that is a compound of the SBS has a protective effect and prevents mitochondrial uncoupling after 24 hour cold storage.
