Essay

Dissertation describes the morphological, phenotypical, functional and immunobiological properties of isolated human fetal liver (FL) cells before and after cryopreservation using the composition of permeable and nonpermeable cryoprotectants. Cryopreservation under protection of cryoprotectant dimethyl sulfoxide (DMSO) in final concentration of 5% using slow cooling rate (1°C per min), storage in liquid nitrogen, thawing at 37°C and washing out the DMSO, allowed to save the cellular composition and immunological properties of FL cells as well as resulted in the increase of number of hematopoietic stem/progenitor cells. At the same time, the decrease in the survival rate and functional activity assessed by Alamar Blue reduction and colony-forming activity was observed. The application of nonpermeable substances, such as PEO-8000 and sucrose as additives to the cryoprotective medium, contained DMSO, allowed to achieve high survival and viability rates of FL cells as well as to reduce the effective concentration of DMSO.
