Essay

The object is the development of technology for obtaining of mouse single-chain antibodies against recombinant human interferon a2b (IFN- a2b) in Escherichia coli; the aim is the achievement of high-efficient expression of ScFv genes in E. coli and the development of manufacturable approach for isolation of target proteins in a pure and functional form; the methods of microbiology, molecular biology, genetic engineering and biochemistry were applied; the novelty lies in an obtaining of highly specific ScFv against human IFN- a2b, development the approach for affinity matrix obtaining using the orienting and non-covalent immobilization of ScFv on cellulose carrier, and obtaining the affinity matrix for one-step purification of recombinant IFN- a2b directly from clarified lysates of producer's cells; results include the development of laboratory technology for obtaining of ScFv against IFN- a2b by expression in E. coli and proposition for their application in purification process for recombinant humanIFN- a2b; the field is biotechnology.
