Bondar N. The chemical and toxicological investigation of amlodipine

The thesis is devoted to chemical and toxicological investigation of calcium channel blocker drug - amlodipine. The results of amlodipine identification by methods TLC, HPLC, UV - spectroscopy are presented and possibility of this methods use in the presence of other cardiovascular drugs has been demonstrated. Accessible methodologies based on the extraction-photometric, UV -spectrophotometric and HPLC methods for amlodipine quantitation have been elaborated. The conditions of amlodipine extraction from water solutions by organic solvents depending on pH medium have been studied. Comparative study of amlodipine isolation effectiveness from a biological material by generally accepted in toxicological analysis methods has been carried out and the own effective methodologie of amlodipine isolation by methanol and acetonitryl (2:3) have been offered. The methodologies of amlodipine isolation from urine and blood have been elaborated. The distribution of drug in organs of poisoned animals and safety in a biological material under putrefaction have been studied. The scheme of amlodipine directional toxicological analysis in a biological material has been offered on the basis of carried out investigations.
