Essay

The Dissertation is aimed to investigate the influence of cryopreservation on survival of human fetal liver (HFL) fibroblast-like precursors. The amount of about 50 % HFL cells derived from primary suspension is capable of plastic attaching. Colony forming efficiency of HFL fibroblast-like precursors was about 0.002-0.003 %. Exposure of HFL cells to 0.7 or 1.4 M dimethyl sulphoxide results in stimulation of osteogenic differentiation induction of HFL fibroblast-like precursors in vitro. Role of overcooling for HFL fibroblast-like precursor survival during cryopreservation has been determined. The initiation of ice-formation during slow freezing of HFL primary suspension with 0.7 M or 1.4 M dimethyl sulphoxide is necessary for protection of the precursors which able to form the colonies and to be differentiated into both osteogenic and adipogenic lineages.
