Object - functioning of the intracellular and intercellular regulatory cells, responsible for T-lymphocytes activation. The aim of the study - to study the role of pttg-knock-out in activation of T-lymphocytes. Methods: biochemical, cell-biological, statistical. It was investigated PTTG-production in murine T-lymphocytes, as well as an effect of pttg-knock-out onto T-lymphocytes blast-transformation. The influence of pttg-knockout onto cell proliferation and c-myc expression was also studied. It was studied the role of pttg-knock-out in apoptosis. The effect of pttg-knock-out onto TGFbeta-1 dependent inhibition of T-lymphocytes activation. By means of proteomics-based approach, it were detected and identified proteins with changed expression in T-lymphocytes without pttg gene comparing to activated wild-type T-lymphocytes . The experimental data, obtained in proteomics-based studies were validated by ELISA, and quantitative data were obtained. Area of use - cell biology, pharmacology.
