Object: interaction of J774.2 macrophages and target cells with different susceptibility to their cytotoxic action. The aim of the study is to investigate the changes occurred in macrophages which interact with target cells. Methods: biochemical, cell-biological, statistical. We have estimated cell viability in mixed cultures of effector and target cells using such criteria as membrane integrity, phosphatidyl serine externalization, decrease in mitochondrial potential, DNA fragmentation and activation of caspases. The influence of different culture condition on cytotoxic action of macrophages towards target cells was studied. Using DNA arrays and RT-PCR method, the expression of genes coding for cytokines and signal transduction pathway elements was investigated in macrophages isolated from cocultures with target cells. DNA-binding activity of transcriptional factor NF- kappaB from nuclear extracts of macrophages cocultured with target cells. Area of use - cell biology, pharmacology.
