Essay

The thesis is devoted to the role of water in the interaction of basic protein compounds with fumed silica surface; influence of different factors on functionality of adsorbed proteins and the nanosilica influence on blood coagulation. Formation process of supramolecular systems containing blood plasma proteins (albumin, immunoglobulin and fibrinogen), water and fumed silica were studied by NMR spectroscopy and thermodynamic parameters of these systems were determined for the first time. It was established that concentration of bound water (Cuw) for all examined proteins increase with protein concentration decreasing and in approach of infinitely diluted solution allows to determine the change of Gibbs free energy, depended on protein molecules self-association (Gas). It was shown that addition of fumed silica to suspension containing immunoglobulin leads to adsorption of protein molecules on silіca surface but in case of fibrinogen - to coagulation. It was revealed that hydration and self-associationof protein molecules during fibrinogen to fibrin transformation decrease as follows fibrinogen>fibrin(monomer)>fibrin(polymer).It was found that interaction of serum albumin with sugars (sucrose, fructose, glucose) leads to significant protein dehydration. In case of sucrose dehydration is equal 90 %.
