Essay

The thesis covers studying of steroid-producing function of testes cells during cryopreservation. In the research the method of obtaining the interstitium cell suspension (ICS) having a substantial steroidogenic potential has been developed. The effect of cryopreservation conditions (cryoprotectant concentration, cell incubation with cryoprotectants prior to freezing, cooling of samples at various temperature intervals) on survival of steroidogenic cells in ICS has been investigated. This resulted in the designing of cryopreservation protocol for ISC, which provided the highest survival and functional activity of steroidogenic cells if compared with the protocols already existing for testicular biological material. In the set of experiments in vivo at allo-transplantation there was also shown that cryopreserved material of testes as ICS according to the program which was presented within this work had a considerable ability to compensate the lack of androgens in comparison with fragments of testis andcontributed to an own hormone-production in an animals organism with orhiectomy and toxic experimental hypogonadism caused by cadmium chloride.
