Essay

The thesis is dedicated to investigating the state of apoptosis spontaneous and induced by different factors ofperipheral blood mononuclear cells (PBMCs) at atopic bronchial asthma. We show that PBMCs from patients with atopicbronchial asthma (ABA) are more resistant to spontaneous apoptosis and exhibit enhanced expression of Bcl-2 proteincompared with healthy donors (HD). The antiapoptotic effect induced by sera from patients with ABA is dependent on high levelexpression of Bcl-2 protein and low level expression of p53 protein on PBMCs from patients with ABA. Cross-linking Fasreceptors by anti-CD95 monoclonal antibodies (mAb) on PBMCs from patients with ABA and HD induced resistance toFas-mediated apoptosis in the presence of serum from patients with ABA. Moreover, expression of р53 protein tended toincrease following activation, but expression of Bcl-2 protein declined. Cross-linking MHC class I molecules by anti-HLA-ABCmAb on PBMCs from patients with ABA and HD in the presence of serum from patients with ABA led to decreased apoptosisand enhanced expression of Bcl-2 protein. Cross-linking MHC class II molecules by anti-HLA-DR mAb on PBMCs frompatients with ABA and HD in the presence of autologic sera decreased apoptosis and enhanced Bcl-2 expression.
