Krivda R. Identification of person in forensic medicine on the basis of PCR-analysis of genomic DNA of bone tissue

The thesis is devoted to questions of identification of person in forensic medicine on the basis of PCR-analysis of genomic DNA of bone tissue. In work it was conducted comparative analysis of basic methods for DNA extraction from bone tissue and its amplification. The methodological aspects of conduct-ing of forensic-medicine molecular-genetic research the object of which there is bone tissue of a different structural type and degree of saving were developed. A number of modifications of basic stages of the genomic DNA extraction from bone tissue of changed (body), exhumed, sceletonized corpses and corpses from fireplace were proposed. The principles and terms of PCR-typing of DNA ex-tracted from bone tissue have been developed. Most optimum bone objects for conducting the identification research were chose. Methodical recommendations for a selection, preparation and terms of storage of bone biological material are offered in work. Practical recommendations for conducting of molecular-genetic forensic-medicine identification examination after bone remains were com-posed. Modified protocols of the DNA extraction from bone tissue and it PCR-typing are successfully applied in practice of forensic-medicine examination of different regions of Ukraine.
