Essay

The object - regenerative processes in mucous membrane of stomach after its experimental damage and using the fraction below 5 kDa from cord blood subjected to low temperature influence and liophilization. The aim - to develop a method of extraction, conditions of storage and biological activity of the cord blood fraction below 5 kDa. Methods: liophilization, cryodestruction, ultrafiltration, gel-chromatography, light, laser scanning microscopy, histological, histochemical, biochemical, clinical. Application of cryodestruction and ultrafiltration allows to extract the fraction below 5 kDa from cord blood, the biological activity of which is higher than the activity of comparator agent actovegin. It has been shown that the cord blood fraction possesses antiulcer activity towards the experimental subchronic stomach ulcer model in rats. The cord blood fraction injections caused a reduction in damage area, an increase in glycosaminoglycans synthesis, promoted recovery of structure of gland layer, led in normalization of biochemical and clinical indices of blood indices and faster recovery of mucus membrane as compared to actovegin. Lyoptilization of the fraction proved to be desirable for one year storage. Besides 6-month storage of the frozen fraction at -80°C is also acceptable. The results can be used for improvement of technologies of biologically active component extraction from natural raw materials, and also for development of new antiulcer preparations.
