Essay

Investigation object: biotechnology of drugs for the treatment and diagnostics of diphtheria, based on the use of advanced methods and approaches. The aim of study: to obtain recombinant scFv-antibodies specific to diphtheria toxin, to study their toxin-neutralization properties and ability to be used in diagnostics. Methods: molecular-biological, immunological, immunochemical, biochemical, working with cultures of eukaryotes. ScFv-antibodies to diphtheria toxin were obtained and described. The immune library of cDNA of immunoglobulin genes of mice immunized with recombinant A and b subunits of diphtheria toxin was designed. Universal approach for detection of scFv-antibodies and their complexes with antigens in was proposed. The ability of scFv and their fusions with staphylococcal protein A neutralize diphtheria toxin was demonstrated. Four antitoxic scFv-antibodies inhibiting toxic effects of diphtheria toxin by preventing its penetration into cells were selected. Obtained scFv-antibodies can be used in diagnostics and therapy of diphtheria. Field of application: biomedical institutions.
