Essay

The dissertation investigates the impact of T-lymphocytes in the development of MCF-7 cells in monolayer (2-D) and spheroid (3-D) models. It's found that T-cells humoral factors stimulated the monolayer culture to in-tensive proliferation, reduced adhesive contacts with the substrate and the fall of sur-vival. Instead spheroid culture under the influence of these factors lost proliferate ca-pacity, increasing its adhesive fraction, and survival remains on the control level. Cytofluorymetry study revealed that under the influence of T-lymphocytes in 3-D model reduced the share of apoptotic cells and cells in S phase. In contrast, the proportion of cells in G0/G1 phase increases the powering. At the same time 2-D cul-ture, in control, there cells redistribution in favor of synthetic phase (52%) and in the presence of T lymphocytes increases the powering proportion of cells in G0/G1 phase. For spheroids control concentration of NO ion benchmarks are higher and were in zone 17,5-18,0 nМ, for monolayer - 8,5-9,0 nM. Cocultivation with T-lymphocytes leads to decreasing overall level of intercellular NO and increased pro-liferate capacity of tumor cells.
