Essay

Objects of study: ECHO virus virulence, genotype and phenes. Objective: Rationale and use of laboratory markers of virus virulence ECHO by examining their phenotypic and genotypic properties. To achieve this goal were the following objectives: Create a collection of current in epidemiological terms ECHO types of viruses that circulate in the territory of Ukraine. Set the fitness of phenotypic markers of virulence poliovirusnyh for ECHO viruses. Find sorption properties of bentonite ECHO viruses, including the type of virus. Assess the potential use of signs rct40 vnutrishnotypovoyi for differentiation of viruses. Identify the method of sequencing fragments of RNA mutational changes in the loci of surface proteins VP1 nucleotide level among strains isolated from healthy and sick individuals. Established a collection of viruses ECHO 3, 6, 11, 13, 24, 30 types circulating in the territory of Ukraine. Proven ability to use bentonite marker for differentiation of virulent strains of ECHO virus. Against strains of ECHO virus 6 and 30 types of fitness set rct40 marker for differentiation of these viruses for virulence. Installed 13 mutations at nucleotides vp1 gene in strains isolated from healthy and sick persons that can be used as a genotypic marker for virulence of ECHO virus type 6, which can be used as a molecular epidemiological marker in monitoring the circulating virus. Moreover, the results of the thesis introduced in the educational process and research work of the department of microbiology, virology and immunology of the National Medical OOBohomolets, department of medical biology, microbiology, virology and immunology of the Ternopil State Medical University Horbachevsky. Has been shown in epidemiological terms of actual types of ECHO viruses that circulate in the territory of Ukraine. Proved that ECHO viruses characterized by dissociation bentonite genetic marker after reproduction of viruses in cell culture with the formation of two populations, one of which sorbuyetsya on bentonite (Abent +), the other - not sorbuyetsya (Abent-). Found that among the studied cell cultures perescheplyuvalnyh RD, HEp-2, Vero greatest vulnerability to viruses ECHO showing line RD. Determined eligibility for the differentiation marker rct40 avirulentnyh and virulent strains of ECHO viruses in cell culture using the HEp-2. Revealed 13 point mutations in the comparative study by sequencing the nucleotide sequence of the gene vp1 strains ECHO type 6, isolated from healthy and sick individuals.
