Konvalyuk I. Gentiana lutea L. and G. pneumonanthe L. tissue and organ culture: generation and molecular-genetical characteristics.

The object - cultivation in vitro of two model gentians' species of Ukrainian flora, investigation genetic variability/stability in vitro culture; the aim is to obtain tissues, organ culture and regenerants of Gentiana lutea and G. pneumonanthe, investigation into their somaclonal variation using molecular-genetic markers; the methods applied - methods of work with tissues and organs culture of plants, RAPD- and ISSR-PCR, DNA electrophoresis in agarose gel, statistical methods; the novelity - for the first time for the species G. lutea and G. pneumonanthe a comprehensive study that includes several stages was conducted: from the selection conditions for a long unorganized growth of tissue culture to direct and indirect organogenesis, receiving and rooting of regenerants, evaluation of the somaclonal variability level of in vitro cultures of these species; the results - conditions for obtaining tissue culture, isolated roots, induction of direct and indirect regeneration of plants G. lutea and G. pneumonanthe was developed, the level of molekular-genetic variability of tissues, organ culture and regenerants was estimated; the branch of knowledge is the biotechnology of plants.
