Essay

The dissertation is devoted to the actual problem of using alternative models to traditional researches on animals for toxicological and hygienic assessment of chemicals. In the presented work there were studied human cell cultures of different organ origin as a model for assessing the toxicity of heavy metal compounds (cells lines A-549, HaCaT, Colo-205, HepG2, 293, IMR-32, U-373, normal fibroblasts). Using methylthiazolyltetrazolium, neutral red, sulforhodamine B assays we found that cytotoxic effects research data of heavy metal compounds in organ-specific human cells cultures corresponded to the acute and organ toxicity data from experiments using animals. In this case, the study of cytotoxic effect of chemicals on human cell lines of various organs can predict the degree of acute and organ toxicity in animals and humans by the indicator EC50 to these cells during toxicological and hygienic evaluation.
