Essay

Object of the research: hematopoietic system of Wistar rats. The aim of the work: to determine the characteristics of hematopoietic system functioning of the rats, irradiated with Strontium-90 radionuclide, at single dose and prolonged administration, and to indicate the most informative criteria for bone marrow hematopoietic function assessment in case of internal irradiation. Methods: cultural, biochemical, light microscopy, statistical. Results: assessment of hematopoietic system functioning was performed in case of prolonged internal exposure of the rats to Strontium-90. Detrimental effects on hematopoietic stem cells and progenitors were revealed using in vitro culture and diffusion capsule in vivo model. The most common effects include inhibition of colony-forming activity of bone marrow hematopoietic progenitor cells in culture conditions, infringement of differentiation and detrimental changes in cell clones, predominant differentiation into eosinophilic lineage, upraise in level of circulating progenitor cells in the peripheral blood. Apart from that, exposure to Strontium-90 induces reduction of the colony-forming activity of stromal bone marrow progenitor cells, reduction of their hematopoietic stimulatory capacity as feeder layer cells, and changes in metabolism of the extracellular matrix, which are indicative of the violation of the hematopoietic microenvironment. Area of use: cell biology, hematology, radiobiology.
