Essay

The thesis is devoted to the obtaining of transgenic carrot (Daucus carota L.) and model tobacco plants (Nicotiana tabacum L.) by means of A. rhizogenes- and A. tumefaciens-mediated transformation. The expression peculiarities of the genes of imunogenic secretory Mycobacterium tuberculosis proteins and recombinant human interferon alpha-2b driven by constitutive 35S CaMV promotor and root-specific Mll sugar beet promotor are discussed. The root-specific accumulation of recombinant biologically active proteins in transgenic plants and hairy root culture of the described plants is shown. The peculiarities of the introduced feature inheritance are analysed for the Т1 generation of the transgenic tobacco plants. Callus, suspension and hairy root cultures initiated from the transgenic carrot plants with the highest level of interferon alpha-2b accumulation were characterized by high level of the recombinant protein antiviral activity for carrot hairy root and suspension cell cultures. Key words: human interferon alpha-2b, secretory Mycobacterium tuberculosis proteins, carrot (Daucus carota L.), Agrobacterium-mediated transformation, tissue-specific transgene expression.
