Essay

The object of study - MGMT gene expression at the protein level in normal and tumor tissues of patients, the role of MGMT in the treatment of malignant tumors; goal - the study of repair enzyme MGMT role in resistance of tumor cells to the cytotoxic effects of alkilating chemotherapeutic drugs; methods - clinical, morphological, molecular-genetic, cultural , statistics; the novelty of results - in the first time data about the features of MGMT expression in II-IV grade glioma samples of patients of Ukrainian population was obtained and summarized; data about the features of MGMT expression in samples of benign heart myxoma obtained and summarized; a correlation analysis between the expression level of the enzyme MGMT in leucocytes, tumor and normal tissue of the same patients was made; in the first time found statistically significant correlation between the level of MGMT expression in samples of leukocytes and tumor tissue of cancer patients: negative correlation for glioma and positive correlation for myxoma; a comprehensive assessment of the dynamics of changes in the cytotoxic effect of alkylating chemotherapeutic drugs depending on their dose and the presence of MGMT in the cells at the primary tumour and standard human cell lines was carried out; the feasibility of using molecular genetic studies of presence of MGMT in cancer samples for the predict of efficacy of alkylating chemotherapy was been experimentally demonstrated; introduced - in the educational process at the Department of Pathological Physiology of M. Gorky Donetsk National Medical University, Kharkiv National Medical University, Lugansk State Medical University, Bukovina State Medical University (Chernivtsi), S.I.
