Varshaver P. The efficacy analysis of cell therapy using mesenchymal stem cells of bone marrow in chronic ischemia treatment of low extremities (experimental-clinical research)

The study object is a chronic ischemia of lower extremities; the purpose is to improve the treatment results of patients with chronic ischemia of lower extremities by substantiation and clinical usage of autologous transplantation of mesenchymal stem cells of bone marrow; the methods include methods of cellular cultivation, МТТ-assay, phase-contrast microscopy, histological, immunohistochemical, instrumental, forced motion test, statistical; the novelty is especially, it was established that the aseptic inflammation induced by the intramuscular introduction of autoblood doesn't result in neoangiogenesis; it was also proved that the usage of mesenchymal stem cells induces neoangiogenesis effect in the ishemic extremity, improves the microcirculatory results of tissue perfusion and rehabilitates the functional state; there were established the parallels between the quantity of intramuscular vessels, fibrous alterations in the muscles as well as the capacity of rats to survive the physical activity; it was proved an effectiveness of intramuscular introduction of cellular transplant in contrast to the systemic intravenous introduction; based on the analysis of autoblood and autologous mesenchymal stem cells implementation for neoangiogenesis stimulation in the ischemic extremity; it was proved in the first time that the aseptic inflammation doesn't result in neoangiogenesis; the results include that the intramuscular introduction of mesenchymal stem cells in patients with III and IV degree of chronic ischemia of lower extremities promoted with the angiogenesis activation processes, that in its turn allowed to decrease the quantity of high amputations in 5-6 times and to increase the quality of life according to the TASK criteria; It is inculcated - in medical practice, in an educational process; Sphere - medical practice.
