Essay

The research object: the impact of culture conditions and cryopreservation on the morphological properties of newborn piglet adrenal cell cultures. The research aim: to investigate the morphological characteristics of newborn piglets adrenal cells during culturing in vitro, and to explore their changes after cryopreservation depending on the initial state of biomaterial (cryopreservation of fresh selected tissue fragments or primary cell cultures). Research methods: cryopreservation, cell culture, immunohistochemical method, radioimmunoassay method, light and fluorescence microscopy, morphometric analysis, standart statistical methods. It is shown that when cultured cells derived from native adrenal tissue fragments in medium containing 10% fetal calf serum, the formation of a monolayer, cytospheres and emerging beta-III-tubulin positive cells are observed. These properties are preserved after cryopreservation of primary cell culture, as well as frozen of tissue sample with cooling rate of 0.3 deg/min. It is also shown that in culture the number of cells expressing 3 beta-hydroxysteroid dehydrogenase gradually reduced without additional stimulation.
