Essay

The research object - the influence of culture conditions and hypothermic storage on morphological and functional properties of the culture of newborn rats' brain cells. The research aim - determination of hypothermic storage effect on the state and behavior of newborn rats' neural brain cells in culture depending on the conditions of microenvironment. Research methods: cultivation, immunocytochemistry, spectrophotometry, gel permeation chromatography, light and fluorescence microscopy, morphometric analysis, methods of statistical analysis. For the first time were carried out comparative studies of the impact of intra- and extracellular culture medium on the effectiveness of hypothermic storage of neural cells in suspension and in the form of multicellular aggregates, as well as the effect of serum and brain/liver extracts on the behavior of neural cells in culture in vitro. It was obtained original data that hypothermic storage of neural cells in the form of aggregates increases the storage time of the cells up to 4 days. The results can be used to design the protocols of hypothermic storage and culturing of neural cells.
