Essay

The research object is the change of the morphological and functional state of the primary suspension of rat adrenal cortex cells (ACC) after incubation and cryopreservation in cryoprotective media based on DMSO, which include and do not include fetal serum (FS). The purpose of the reseach - to determine the influence of some factors and cryopreservation conditions on morphological and functional properties of the rat ACC suspension. Methods: culturing, cryopreservation, radioimmunoassay, colorimetric assay, flow cytometry, volumometry and histochemical methods, light, fluorescence, FRET-microscopy, statistical analysis. It was shown the increase of the number of functionally active cryopreserved ACC by adding of 25% FS to the cryoprotective medium wich include 10 - 15%DMSO. It have been improved the removal step of the cryopreserved media included DMSO and 25% FS by adding FS to the washing medium in the same concentration. It was proposed the usage of such fluorescence microscopy technique as FRET - microscopy to determine the relative position of mitochondria and lipid droplets in isotonic and hypertonic solutions, as well as the potential measurement techniques on mitochondrial membrane with a fluorescent probe JC-1 to assess the steroidogenic capacity of ACC.
