Essay

The thesis is devoted to optimization of Agrobacterium-mediated transformation of bread wheat Triticum aestivum in vitro and in planta, and obtaining transgenic plants carrying genes for herbicide resistance. The correlation between growth regulators (auxin like synthetic growth regulators - dicamba, picloram and cytokinin - 6 benzylaminopurine) that are necessary for the formation of active cells in the meristematic callus and subsequent regeneration of shoots and roots formation of wheat were established. The culture media components increasing the rate of regeneration of wheat and speeding up transformation were developed. Antibiotics concentration of B-lactam group timentin and ceftriaxone, which eliminated A. tumefaciens cells of strain ABI and GV3101 were established. Besides the effect of ceftriaxone on morphogenetic processes in bread wheat was studied. The transformants of Triticum aestivum cultivars Podolyanka and Zymoyarka resistant to herbicyde phosphinothricin were received. The application of ceftriaxone as a growth regulator of wheat was proved. Stimulating morphogenetic processes had place once ceftriaxone was introduced into hormone free culture medium. The improved biotechnique and the first genetically engineered plants of Triticum aestivum cultivars Podolyanka and Zymoyarka resistant to herbicide phosphinothricin through Agrobacterium-mediated transformation in planta and in vitro were obtained.
