Essay

Enzymatic oxidation of polyunsaturated fatty acids by lipoxygenase. Establishment of laws of manifestation of the functional activity of 9- and 13- lipoxygenases using natural compounds and factors - plant hormones, primary lipoxygenase products, abiotic stressors. RP-HPLC, chromatographic methods of purification of proteins and lipids, UV spectrophotometry, enzymatic synthesis, kinetic and mathematical methods. The significant changes in the activity of plant 9- and 13-lipoxygenase at low temperature, high salt concentration and under exogenous 24-epibrassinolide and abscisic acid influence were founded. In model systems of mechanical wounding on potato tubers under conditions of exogenous linoleic acid 13-hydroperoxide or arachidonic acid 15-hydroperoxide application was found a significant decrease in 9-lipoxygenase activity. It was determined that introduction to the structure of 24-epibrassinolide additional active ingredient in the form of balance heteroauxin decreased the IC50 on the order in inhibition of potato 9-lipoxygenase. It was determined the activatory effect of lysophosphatidylinositol and lysophosphatidic acid on oxidation of linoleic acid by potato 9-lipoxygenase. Field of use - bioorganic chemistry, biochemistry, enzymology and biotechnology.
