Essay

Object of investigation: trivalent influenza vaccines and experimental samples of newly created liposomal viral vaccines. Research objective: to increase immunogenicity of the split influenza vaccine by modifying the adjuvant and antigenic components, preserving its safety. Methods of research: immunological, biochemical, microscopic, physicochemical, mathematical and statistical. Equipment: Bioanalyzer "Agilent-2100" (Agilent Technologies, USA); Atomic Force Microscope ACM NT-206 (Belarus); Microscope Primo Star ILED (Zeiss, Germany). For the first time the complex of measures was developed to optimize the composition of influenza vaccines with the simultaneous modification of antigenic and adjuvant components, which in combination with immunogenicity increase provides antigenic load reduction, allowing them to be used in different age groups and is economically significant. For the first time the obtained data extends the concept of specific antibody and vaccine protein profile production, quantitative contents and protein molecular weight in its composition, role of liposomal component surface charge and its ultrastructure in the process of complete response formation and immune memory for liposomal viral vaccines, proven high safety of influenza vaccine modified with cationic liposomal compositions based on phosphatidylcholine. Results of the research may serve as justification for preparation of highly immunogenic safe liposomal influenza vaccine for immunological prevention for all age groups. Method, developed for liposomal vaccines immunogenicity increase is patented (Patent of Ukraine for Utility Model No. 108844). Use of high-resolution noncontact atomic force microscopy aimed at visualization of liposomal vesicles surface ultrastructure in the process of liposomal vaccines preparation. Degree of implementation: 3 acts of introduction, 1 patent, 1 newsletter. Area (industry) of use: public health (medicine: immunology, vaccine).
