Konup A. Diagnosis of Viral Diseases of Grapes When Creating Certified Non-Virus Planting Material of Grapes in Ukraine

The dissertation is devoted to the actual problem in the field of viticulture, namely: improvement of diagnostic methods for obtaining a virulent (healthy) grapevine planting material in Ukraine, as well as recommendations for the production of quality grape plants. The actuality of the work was conditioned, first of all, by the need for the diagnosis of virus diseases of the grapes caused by harmful viruses, such as: Grapevine Leaf Roll-Associated Virus (1–9) (GLRaV1-9), Grapevine Fanleaf Virus (GFLV), Grapevine Fleck Virus (GFkV), Virus A (Grapevine Virus A (GVA) and Grapevine Virus B (GVB) Rugose wood – associated viruses. Phytosanitary examination for the presence of external symptoms of viral diseases was carried out at industrial vineyards of South of Ukraine. The scientific novelty of the obtained results is that for the first time in Ukraine, the method of identification of grape viruses using polymerase chain reaction real-time mode with hybridization-fluorescence detection using Ukrainian isolates is used. Real time polymerase chain reaction method allows the identification of different grape viruses by means of unique pairs of primers in a short time. In the process of diagnosis, the same basis, except for specific primers and fluorescently labeled DNA probes, was used for the polymerase chain reaction products, and the composition of the polymerase chain reaction reaction mixture. A comparative analysis of the diagnosis by the method of immuno-enzymatic analysis and the polymerase chain reaction method in real time with hybridizationfluorescence detection proved the undoubted advantage of the latter in the economic aspect and let as get to reliable results.
