Essay

The thesis is dedicated to assessment of individual conditions for MSCs cryopreservation and formation of 3D scaffolds using human blood plasma-based solutions. It was established that plasma-based hydrogels ensured adhesion and proliferation of MSCs and could be used as 3D carriers. The method of MSCs cryopreservation, including pretreatment with sucrose, was improved by replacement of culture media with blood plasma in cryoprotective solution. After cryopreservation in human blood plasma, supplemented with 0.2M sucrose and 1% DMSO (PS1D solution), MSCs had high viability and metabolic activity values. PS1D solution could form a stable hydrogel allowing creation of bioengineered structures. When applied on the surface of fullthickness wounds as a part of hydrogel PS1D, cryopreserved MSCs improved wound 23 healing. GFP+ - MSCs in PS1D hydrogel were visualized within the wound defect area for 5 days after application.
