Objects – medical substances sildenafil and vardenafil; aim −to develop a set of methods to effectively extract, identify and quantify sildenafil, vardenafil and their metabolites in biological samples for the forensic chemistry or clinical and toxicological practice in cases of lethal and acute combined poisonings diagnostic; methods – color and precipitation reactions, thin layer chromatography, UV spectrometry, fluorimetry, gas chromatography-mass spectrometry, high-performance liquid chromatography with UV-, fluorescence- and mass- detection; ultra-high performance liquid chromatography with UV- and mass- detection; results − the step-by-step method of chemical and toxicological study of biological material for the presence of sildenafil, vardenafil and their metabolites in combined poisoning cases has been developed; to detect sildenafil and vardenafil, a number of sedimentary and color reactions were proposed, analyzed the thin layer chromatography solvent systems; described conditions for sildenafil and vardenafil detection and quantification using UV spectrophotometry; fluorimetry; gas chromatography-mass spectrometry, high performance liquid chromatography with mass-, UV- and fluorescence- detection; ultra-high performance liquid chromatography with mass and UV- detection; the conditions of sildenafil and vardenafil extraction from aqueous solutions and solid-phase extraction using Oasis HLB cartridges and with H-clinoptilolite were studied; effective methods of the investigated compounds isolation from biological materials were proposed; the developed methods for sildenafil and vardenafil detection using gas chromatography and high performance liquid chromatography-mass spectrometry in biological matrices were validated; the distribution of sildenafil, vardenafil and their metabolites, in organs and biological fluids of rats 3h and 24h after co-administration with nitroglycerin was investigated using mass spectrometry; introduced − into the practical activities of the Main Bureau of Forensic Medical Examination of Ministry of the Health of Ukraine and the Odessa Regional Bureau of Forensic Medical Examination, into the scientific work and educational process of higher educational institutions of Ukraine; branch - pharmacy.
