Essay

The object of the study was the process of wound healing in hairless rats after skin cryodestruction. The subject of the study - destructive and repair processes in the tissues of hairless rats after skin cryoablation and the introduction of a extract of cryopreserved piglet`s skin fragments or an aqueous colloidal solution of unmodified fullerene C60. The aim of the work was to determine the сharacteristics of the wound healing process after skin cryodestruction in hairless rats after introduction of the extract of cryopreserved piglet`s skin fragments and aqueous colloidal solution of C60 fullerene. The following methods were used in the work: cryosurgical, contact thermometry, clinical examination (including planimetric), laboratory (hematological, biochemical), histological (morphometric, immunohistochemical); spectrophotometric, statistical. The experimental model of the destruction and regeneration processes after skin cryoablation in hairless rats was presented, which was characterized with local and systemic manifestations of destruction and inflammation and could be used to study the processes occurring after cryosurgical treatment of tumors. It is shown that the introduction of the extract of cryopreserved piglet`s skin fragments (PSCFE) or aqueous colloidal solution of C60 fullerene (AQSUF C60) was accompanied by accelerated wound healing after skin cryoablation, manifested by a decrease in morphological manifestations of destruction and inflammation as well as increased manifestations of reparative processes at macro- and microscopic levels. It was demonstrated that the administration of the PSCFE or AQSUF C60 after skin cryoablation in hairless rats contributed to a decrease of markers of destructive and inflammatory processes, manifested by normalization of prooxidant-antioxidant balance. It has been shown for the first time that tissue regeneration by skin cryodestruction after PSCFE administration is more organotypic than after AQSUF C60 one, which is manifested by the formation of skin appendages and greater balance of apoptosis and proliferation processes. For the first time it was found that the use of PSCFE and AQSUF C60 in animals after cryodestruction of the skin reduced the level of markers in the serum of destructive and inflammatory processes. Based on the data of studying the influence of AQSUF C60 on the processes of lipid peroxidation and morphological indices of destructive and regenerative processes in tissues after skin cryoablation, a method of wound healing was developed (patent of Ukraine UA111022 U). The obtained data on the stimulation of cell proliferation by fullerenes on the background of a decrease in their apoptotic activity can experimentally justify the feasibility of performing studies of the long-term effects when using the AQSUF C60 in various experimental models. The findings can be an experimental justification for the feasibility of research on the possibility of including PSCFE into wound treatment protocols.
