Essay

Polymethinecyanines, styrylcyanines, polymethine chalcones. Metallocomplexes - phthalocyanines with out of plane ligands. Albumins (human, bovine, horse, from egg), globular proteins - imunoglobuline G, bovin carbonanhydrase (BCA), native fibrilogenic proteins- ?-synuclein, insulin, ?-lactoglobulin, lysozime and their amyloid aggregates, oligomeric ?-synuclein. Absorption spectroscopy in UV and visible regions, fluorescent spectroscopy, fluorescent polarization spectroscopy, fluorescent time-resolved spectroscopy, atomic force spectroscopy, gel-electrophoresis, computer simulation, replacement fluorescent titration, microfluid chromatography, two-photon excited fluorescent spectroscopy, MALDI-TOF mass-spectrometry. It is shown that mono and trimethine cyanine dyes are efficient probes for the amyloid fibril detection, when tri and pentamethine cyanine dyes for detection of oligomeric aggregates. The spatial model of complex of monomethine cyanine dye dye with binding channel of amyloid fibril is proposed. The high inhibitory activity of series of phthalocyanines with out-of-plane ligands in reaction of insulin fibril formation was shown. The high specificity of squaraine dyes with long N-alkyl substituents to albumines was discovered and for the most efficient squaraine dyes the affinity to binding sites of human serum albumin was studied. The applicability of pentamethine cyanine dyes as probes on native globular protein, their partial denatured conformation and monitoring of conformational transitions was shown. Series of fluorescent probes: for detection of amyloid fibrils and monitoring of fibrilogenesis reaction, for post-electrophoretic visualization of proteins on polyacylamide gels, for non-specific protein detection in microfluid systems were developed and adopted for market.
