Essay

Object of the study: development of malignant and benign thyroid tumors. Aim: to establish the role of oncogene in modulation of both, receptor tyrosine kinases and thyroid specific proteins expression, and to reveal there complex effect on development, morphogenesis and the phenotype of papillary thyroid carcinomas (PTC). Methods and equipment: histological, immunohistochemical, molecular biological, statistic; microscope, thermocycler, thermostat, fridge. Theoretical and practical results: The multifocality of the malignant transformation of thyroid cells with the establishment that the induction of oncogenes is associated with unbalanced increasing of transcriptional activity of different tyrosine kinases, decreasing NIS and aggressive phenotype of PTC was been demonstrated. The differences in realization of oncogenic effects in benign and malignant thyroid tumors were been discovered. The new method for detection of rearranged oncogenes (RO) without there type usinf real-time PCR was proposed. It was shown that both, NIS-negative PTC and PTC with the induction of RO are the most aggressive by invasive characteristics, have worth prognosis with a high risk of development distant metastases. Novelty: The interrelation between induction of oncogenes, activity of RTK system, expression of TSP, and the phenotype of PTC has been demonstrated. In first the differences in pathways of oncogenic regulation in benign and malignant thyroid tumors were shown. The dynamics of molecular biological characteristics of PTC according the patients age was shown. The possibility to identify the basal level of NIS expression in thyroid tumors has been shown in first and the using of NIS-negativity as a marker of PTC aggressiveness has grounded.
