Object of research - the effect of cryopreservation, two-dimensional and three-dimensional culture on the properties of multipotent mesenchymal stromal cells from different sources. Aim - the establishment of the basic regularities of the implementation of morphological and functional properties of multipotent mesenchymal stromal cells from different human tissues during in vitro culture and the development of new approaches to their cryopreservation and three-dimensional and cultivation. Methods - in vitro cell culture; programmable freezing; flow cytometry, cytochemistry, immunocytochemistry, PCR, spectrophotometry, fluorescence spectroscopy, light, fluorescent and scanning electron microscopy, morphometric analysis, statistical analysis of the results. For the first time, a comparative study of morphological and functional properties of MSCs from different human tissues during monolayer and three-dimensional culture within three-dimensional carriers, different in structure, shape and physical-chemical properties. For the first time it was shown that the presence of sucrose in the culture medium increases cells' resistance to further cryopreservation in case of the presence of sucrose in the cryoprotective medium. For the first time, the possibility of cryopreservation of cells within wide-porous cryogel scaffolds was proved and the effect of cells' spreading on the surface of the carrier on their cryodamage was demonstrated.
