Essay

The thesis deals the creation of a systematic approach to the development of methods for the chemical-toxicological analysis of antidepressant action drugs and the development of the algorithm for carrying out the toxicological studies of the biological samples for the presence of antidepressants, appropriate for use in forensic and toxicological examination, drug-testing and clinical toxicology. The isolation recovery of 15 antidepressants from the biological material by the general isolation methods of drugs has been determined. The special method of antidepressant isolation by chloroform followed by the extraction purification using system of non-aqueous solvents such as n-hexane – acetonitrile has been developed considering the lipophilic properties of the antidepressants. Optimal conditions for the extraction of antidepressants from blood and urine by the liquid-liquid extraction method have been determined. The TLC-screening scheme of antidepressants which was able to distinguish all drugs being studied has been developed. Methods for antidepressant identification and determination in the biological samples by HPLC-UVD, UV- and extraction spectrophotometry have been developed. Methods for detection of antidepressant metabolites in urine have been developed. The structures of antidepressant metabolites have been established by mass-spectrometry method.
