Essay

The thesis is devoted to the investigation of the influence of cryopreserved multipotent mesenchymal stromal cells (MMSCs) of rats obtained from different sources (fat and cartilage tissues) under local and systemic administration on the dynamics of the experimental arthritis course. The task of therapy of rheumatological diseases is relevant worldwide. According to the WHO, in the developed countries of the world, inflammatory diseases of the joints and connective tissue take the second place after cardiovascular pathology among the causes that lead to disability and death of patients. Arthritis is characterized by chronic inflammatory synovitis and progressive joint destruction, leading to severe disease and disability. There are many etiological factors for the RA development, as for this pathology therapy, recently more and more attention has been paid to the possibilities and effectiveness of the use of multipotent mesenchymal stromal cells (MMSCs). The research object was the course of pathological changes in the homeostasis system and ankle joints of rats on the background of experimental arthritis, namely adjuvant arthritis (AA). The research subject was the biological properties of cryopreserved multipotent mesenchymal stromal cells obtained from adipose and cartilage tissues. The research task was as follows: to investigate the effect of cryopreservation on the phenotype and functional properties of MMSCs derived from adipose and cartilage tissues; to determine the timing of the development of AA in rats (acute and chronic phases) based on the study of the dynamics of degenerative changes in cartilage tissue and indices of inflammatory markers in the blood of rats; to evaluate the effect of MMSCs on the state of cartilage tissue in rats with AA; to determine the role of MMSCs from different sources in correcting some indices of cell immunity in the rats with AA; to assess the influence of CrMMSCs from different sources on the dynamics of changes in inflammatory markers in the blood of the rats with AA. Materials and methods: pathophysiological, cryobiological, cytofluorimetric and morphometric, histological and morphometric, immune histochemical, immune cytofluorimetric, biochemical, methods of statistical processing of the obtained results. The dissertation presents a theoretical generalization and a new experimental solution to a scientific task aimed at correcting the consequences of arthritis and activating the processes that contribute to the restoration of the homeostasis system as well as the structural and functional properties of damaged articular cartilage due to the local and systemic administration of cryopreserved MMSCs obtained from adipose and cartilage tissues . The research findings can be used to justify and develop the methods of treatment of arthritis in clinical practice.
