Essay

The dissertation covers the study of the impact of administration of cryoextract and secretomes from intact and cryopreserved cultures from the dorsal root ganglia on the reproductive system of female rats of late reproductive age. The experimental results allowed us to conclude that the cryoextract and secretomes from the intact and cryopreserved cell cultures from DRG were powerful modulators of folliculogenesis in late reproductive age ovaries. Administration of secretomes to late reproductive age animals increased the average number of estrus phases and decreased the total number of follicles of hormonally independent stages of development (primordial and primary follicles). At the same time, an increase in the number of follicles at the stage of atresia was observed, which indicated that as a result of the activation of primordial follicles and their further development, an excess of antral follicles appeared, which subsequently underwent atresia. In animals of late reproductive age, there was a 6.5-fold increase in the LH/FSH index after administration of intact culture secretome and 14 times after administration of cryopreserved culture secretome. The administration of cryoextract and secretomes had a stimulating effect on the uterine contractile activity of late reproductive age rats. The activating effect was more pronounced when secretome from cryopreserved cell culture was used, where spontaneous contractile activity (by 1.2 times), and OT-induced (by 1.2 times) as well as KCl-induced (by1.3 times) contractile activity increased. The effect of increasing contractile activity of the uterus occurred due to increased expression of myometrial smooth muscle actin, which was determined by immunohistochemistry. At the experiments on the fertilization of rats, it was established that intact late reproductive age animals did not produce offspring, while in groups of the same age animals with the introduction of secretomes from cell cultures from dorsal root ganglia, the average fertility of animals was 66-70%. The findings indicate the effectiveness of using dorsal root ganglia-derived cell culture secretomes and neurotrophic factors obtained to correct the age-related involutional changes in the female reproductive system. In general, the effectiveness of the biological impact of the compositions used on all the studied indices of the reproductive system of female rats can be distributed in the following order: cryoextract secretome from an intact culture secretome from a cryopreserved culture.
