Essay

Dissertation work is dedicate to research of latent virus of hop and creation of modern effective test-system for identification of this virus by the method of RT PCR and also possibilities of application of microsatellite markers for genetic differentiation of genotypes of hop (Humulus lupulus L.). Research from the study of biological properties is conducted of the Ukrainian isolate of latent virus of hop with a subsequent removing, cleaning and concentration, and also got electron-microscopic information about this virus. On the basis of the got positive controls with the use of world base of these genetic sequences (GenBank) we developed universal PCR test-system which is directed on a detection and diagnostics of the latent virus of hop. Created PCR test-system allowed conducting screening of the newest sorts of hop of the Ukrainian selection and identify the investigated virus. On the basis of determined sequences of nucleotide developed the system of diagnostics which is based on the using of the RT-PCR with the using of the synthesized markers. The method of identification and differentiation of genotypes of hop of the Ukrainian selection is developed, with the using of SSR-PCR. For development of the molecular test-system we made a genetic analysis of 13 sorts of hop of the Ukrainian selection. It was selected and analyzed 10 microsatellite locus and SSR-primers were synthesized. Standardization of PLR is made and it is adapted to the Ukrainian genotypes of hop. On the basis of findings the computer "genetic" bank of data is created and phyllogenetic links are built between these sorts of hop.
