Essay

The subject of the study is in vitro cultivation of isolated meristem derived from Vitis vinifera L. The goal of the study is to in grape morphogeny during in vitro cultivation of isolated meristem together with developing methods of the clonal micropropagation and improving material planted for establishing virus-free plantations in the Crimea. Methods of the study: cultivation of isolated cells and organs in vitro; biometrics, mathematical statistics. Results of the study: specific traits of the morphogenetical potential in grapes' apical meristem derived from outcast and main gemmae are revealed; MCa culture medium is modified for main gemma's apical meristem cultivation; a possibility of repeated use of microcuttings for increasing of propagation rate is revealed; for adaptation of microplants to in vivo conditions, a two-layer absorbent TERAWET contained substrate is worked out and applied for the first time. A developed technology of the grape clonal micropropagation is recommended for extensive use aiming to obtain planting material of recommended and new varieties of Vitis vinifera, pathogen-free plantations and speeding up propagation.
